The authentication and quality control of medicinal plants are essential for ensuring the safety, efficacy, and consistency of herbal medicines. Traditional pharmacognostic techniques, including macroscopic and microscopic examination, have been complemented and increasingly replaced by modern analytical approaches that offer enhanced specificity, sensitivity, and reproducibility. This review critically evaluates contemporary methodologies for pharmacognostic identification and quality control, with particular emphasis on DNA barcoding, chemical fingerprinting, and metabolomics. DNA barcoding enables the accurate identification of plant species, even from processed materials, and has been successfully applied to authenticate commercial herbal products. Chemical fingerprinting techniques, including HPLC, GC-MS, and FTIR, provide comprehensive chemical profiles that can distinguish authentic materials from adulterants and establish quality standards. Metabolomic approaches offer holistic chemical characterization, enabling the simultaneous analysis of multiple bioactive compounds and the detection of batch-to-batch variations. Despite significant technological advances, challenges persist, including the development of comprehensive reference databases, the establishment of quality thresholds, and the integration of these approaches into regulatory frameworks. This review synthesizes recent evidence, evaluates the strengths and limitations of various approaches, and proposes a multi-tiered quality control framework for medicinal plant authentication.
